Examining spores under magnification — their size, shape, color, and surface detail — is the heart of mushroom taxonomy, and the primary, legal use of a spore specimen. With a decent microscope and a few basics, it's very approachable.

What You'll Need
A compound microscope — ideally one that reaches 400x, and 1000x with oil immersion for fine detail.
Glass slides & coverslips — clean, so stray dust doesn't clutter the view.
Your specimen — a spore syringe, or a spore print plus a drop of sterile water to lift spores from it.
Optional: immersion oil for 1000x, lens tissue, and a simple stain for extra contrast. Browse mycology supplies.
Step by Step
Prepare the slide
Place a small drop from your spore syringe onto the center of a clean slide. Working from a print? Add a drop of sterile water, then lightly touch it to the print to lift some spores.
Add a coverslip
Lower a coverslip onto the drop at a slight angle and let it settle — this spreads the sample thin and helps avoid trapped air bubbles.
Start at low magnification
Begin with the lowest objective (around 40x–100x) to find the spores and bring them to the center of your field of view.
Work your way up
Switch to 400x for a detailed look. For the finest structure, use 1000x with a drop of immersion oil on the coverslip.
Focus with care
Use coarse focus to get close, then fine focus for the sharp detail. Spores are tiny — small, patient adjustments win.
Observe & record
Note the size, shape, and color, and sketch or photograph what you see. This record is the basis of taxonomy work.
What to Look For
The features that matter most for identification are a spore's size, shape, color, and surface ornamentation. Are they round or ellipsoid? Smooth or warty? Thick-walled? Do they carry a small pore at one end? These details are how mycologists tell species apart.

A few tips for cleaner views
• Keep slides, coverslips, and lenses clean — dust and fingerprints muddy the image.
• Adjust the condenser and diaphragm to control contrast; too much light washes fine detail out.
• Don't over-load the drop — a thin, sparse sample is far easier to read than a crowded one.
• A light stain can add contrast to pale spores, making shape and ornamentation pop.

What magnification do I need to see spores?
You can spot them at 400x, but 1000x with oil immersion is where fine detail — shape, wall thickness, ornamentation — really comes through.
Do I need to stain spores?
Not necessarily — many spores are visible unstained. A light stain simply adds contrast to pale specimens, making features easier to read.
Can I use spores from a print instead of a syringe?
Yes. Add a drop of sterile water to the slide and lightly touch it to the print to lift spores into the drop, then cover and view as usual.
Why does my view look cloudy or crowded?
Usually too much sample or dirty optics. Use a smaller, thinner drop and clean your slide, coverslip, and lenses for a clearer field.
