Giant Puffball, Calvatia gigantea, is a soil-dwelling fungus known for producing enormous white fruiting bodies without a traditional cap, stem, or exposed gills. Your syringe contains 10 cc of actively growing Giant Puffball liquid culture, intended for advanced mycological projects.
Giant Puffball does not follow the standard process for Lion's Mane, Reishi, oysters, or other block-grown gourmet mushrooms — it has not been reliably fruited indoors from grain bags, Master's Mix, or sawdust blocks. The most successful published method expanded the culture into sterilized soil spawn, buried it in an undisturbed outdoor grass-covered plot, and let the fungus establish naturally across several growing seasons, relying on seasonal moisture and temperature to initiate fruiting. This Is a Multi-Year Project — and It May Never Fruit
In the documented field trial, Giant Puffball first produced a mushroom after four years, near the end of the fifth outdoor growing season — and only one of two identically inoculated plots fruited at all. Successful mycelial establishment is achievable, but fruiting is experimental and cannot be guaranteed. No mushroom in year one, two, or three does not mean failure — and a well-prepared, fully colonized bed may still never fruit.

1Should I Use Grain or Soil Spawn?
The most dependable published method used sterilized soil spawn, not grain spawn. Researchers filled 0.75-liter jars with ~0.6 L of field soil at 30% moisture, sterilized it, inoculated with Giant Puffball culture, and incubated for six weeks at 77°F before burying it outdoors. That soil-spawn method eventually produced three genetically verified Giant Puffball fruiting bodies.
Grain can support experimental culture expansion, but grain-to-indoor-block fruiting has not been established in reproducible research. For the method with the strongest evidence behind it, we recommend expanding the liquid culture directly into sterile soil.
Recommended soil
Clean soil that is loamy or clay-loam · moderately fertile · able to hold moisture without waterlogging · free from herbicides, fungicides, and chemical treatments · collected from the future growing site when practical · free from large stones, roots, and undecomposed wood. The successful field plot used non-tilled clay-loam beneath mown grass in a partially shaded apple orchard. Giant Puffball is strongly associated with nutrient-rich soil and moderate moisture, but available research does not support a narrow required pH range.
Pro Tip
- Using soil from the future bed gives the culture a familiar mineral and texture profile.
- Don't use heavily fertilized commercial potting soil or soil with synthetic slow-release fertilizer pellets.
- Don't use straight compost, manure, coco coir, or Master's Mix as the spawn medium.
- The soil should stay aerated after hydration rather than forming a dense muddy mass.
2Preparing Soil Spawn
The published method used ~0.6 L of moist soil per 0.75-liter container — roughly 2½ cups of soil in a three-cup container, leaving headspace for filtered gas exchange.
Moisture target (~30% by weight)
Prepared soil should feel evenly damp throughout, hold together lightly when squeezed, break apart when touched, release no free water, and contain no muddy or saturated pockets. The goal is moist, aerated soil — not dry dust and not garden mud.
Preparation steps
- Collect the soil and remove stones, large roots, insects, and plant material.
- Break apart compacted pieces.
- Add water gradually while mixing; confirm even distribution.
- Load ~0.6 L into each cultivation jar, leaving headspace above the soil.
- Install a filtered lid with a self-healing injection port; cover the lid with foil.
- Place the containers in the pressure cooker and sterilize at 15 PSI.
- Let the cooker return to zero pressure naturally and cool completely before inoculation.
The published experiment sterilized its 0.6-L soil containers at 121°C (250°F) for 25 minutes. Because home cookers, jar shapes, soil density, and loads vary, 45–60 minutes at 15 PSI is a more conservative home starting point — a practical adaptation, not the exact published cycle.
Pro Tip
- Don't compact the soil tightly inside the jar; keep the filter dry during sterilization.
- Never inject culture into soil that still feels warm (cooling overnight is recommended).
- Prepare multiple containers — a single small jar provides very little inoculum for a permanent outdoor bed.
3How Much Liquid Culture Should I Use?
A practical starting rate is ~2–3 cc per 0.6-liter soil-spawn jar. A 10 cc syringe prepares ~three jars at ~3 cc each, or four smaller jars at ~2–2½ cc each — enough to distribute the culture through several parts of the soil without making the container excessively wet.
The field study began from agar rather than a commercial syringe, so it didn't establish a standardized LC volume. For a meaningful outdoor trial, consider preparing multiple syringes or expanding across enough jars to make ~2–4 liters of finished soil spawn. Don't empty the whole syringe into one location.
4How Do I Inoculate the Soil Spawn?
Use a still-air box, laminar flow hood, or another clean workspace with minimal moving air.
- Put on clean gloves and wipe the work surface with 70% isopropyl alcohol.
- Clean the exterior of each soil-spawn jar.
- Shake the Giant Puffball liquid culture syringe thoroughly and attach the sterile needle.
- Wipe the injection port with an alcohol swab and let it evaporate.
- Insert the needle through the port and inject ~2–3 cc.
- Direct small amounts toward several areas of the soil.
- Remove the needle, label with the species and inoculation date, and place it in the incubation area.
Pro Tip
- Don't empty the entire syringe into one location, and don't add enough liquid to create a wet pool at the bottom.
- Don't open the container after sterilization.
- A gentle tap or rotation is fine, but don't shake the soil into a compact mass.
- Never flame a needle near freshly applied isopropyl alcohol.
5Soil-Spawn Colonization
Maintain the inoculated jars at:
- Temperature: 75–79°F (best ~77°F)
- Light: darkness or low indirect room light
- External humidity: not important while jars are sealed
- Fresh air: passive filtered gas exchange
- Expected colonization: ~six weeks
In the trial, Giant Puffball colonized the full 0.75-L soil vessel within six weeks at 77°F. On agar the isolates grew only ~4 mm/week, so expect slow, deliberate growth — not oyster-like speed.
Healthy mycelium may appear pure white when young, fine and delicate, cream with age, pale gray or ash gray in colonized soil, organized into visible cords or false rhizomorphs, irregular rather than uniformly circular, and closely associated with individual soil particles. Researchers described plate cultures as white with a light-brown reverse, becoming cream or light brown with age; finished soil spawn appeared light gray and broke apart easily. Pale gray growth is therefore not automatically contamination.
Should I break and shake? A traditional grain-style break and shake is not recommended. If the mycelium is growing only in one area after several weeks, gently turn or tap the sealed jar once to create additional contact — don't repeatedly mix it. The study broke apart intact colonized soil particles only when transferring outdoors.
Watch for contamination
Bright green powdery growth · black mold · pink or red colonies · slimy soil · cloudy liquid · strong sour/fermented/rotten odors · growth that rapidly overtakes the container while the Giant Puffball stays stalled. Do not bury visibly contaminated spawn in the outdoor bed.
6When Is the Soil Spawn Ready?
The soil spawn is ready when:
~Six weeks have passed · mycelium is visible throughout the soil · pale white or ash-gray cords connect different areas · the soil breaks into colonized pieces · no wet uncolonized pockets remain · no green, black, pink, or bacterial growth is visible.
The mycelium may be hard to see in darker soil, and a jar doesn't need to turn solid white like an oyster block. Look for fine gray-white growth, hyphal cords, and colonized particles distributed through the soil.
7Choosing an Outdoor Site
Site choice may be more important than any single laboratory step. The successful research site was a grass-covered apple orchard, ~50% shaded by canopy, on clay-loam soil, previously cultivated but no longer tilled, mowed ~twice per year, protected from disturbance, and naturally exposed to rainfall and seasonal temperatures.
Recommended home location
A lawn edge · an old orchard · a lightly shaded meadow · a grassy garden margin · a pasture edge · a partially shaded area beneath widely spaced deciduous trees. The site should receive natural light but not bake in intense afternoon sun.
Soil conditions
Loamy or clay-loam · moderately rich · moisture-retentive · well aerated · free from standing water · undisturbed by regular digging/tilling · covered with grass or low vegetation. Giant Puffball is a soil saprotroph, not a mycorrhizal species — in the trial, dense mycelium developed through the soil but did not colonize nearby grass or apple-tree roots (so no host tree is needed).
Avoid
Raised beds replaced each season · areas treated with fungicides · frequently tilled vegetable gardens · waterlogged depressions · extremely dry sandy ground · solid compacted clay · high-traffic lawns · locations directly beside roads, industrial sites, or contaminated soil.
Giant Puffball can accumulate certain metals from the soil, so site cleanliness matters especially if any future mushrooms may be considered for culinary use.
8When Should I Establish the Bed?
The documented plot was inoculated on March 30, shortly after the start of the growing season — so early spring is the most research-aligned planting period. Establish the bed after frozen soil has thawed, before hot dry summer conditions begin, when natural rainfall can support establishment, and while the soil is moist but workable.
Early fall may also allow soil colonization before winter in mild climates, but the published success used spring inoculation.
9How Much Soil Spawn Do I Need?
The research plot was ~1 m × 3 m (~32 sq ft) and received ten separate 0.6-L portions of mixed soil spawn — ~6 liters total, or roughly 0.2 L of soil spawn per square foot.
That's ~3 liters for a 4×4-ft bed, or ~6 liters for a 4×8-ft bed. For a home project, use ~2–4 liters for a 4×4-ft bed.
More inoculation points may improve the chance that at least part of the culture establishes. Using only one small jar across a large lawn spreads the culture too thinly.
10Preparing the Outdoor Bed
- Mark the perimeter clearly.
- Mow or trim the grass if necessary.
- Remove thick surface thatch without stripping away all vegetation.
- Loosen only the upper few inches when the soil is heavily compacted.
- Do not completely replace the native soil.
- Water lightly if the ground is dry, and allow excess to drain before inoculation.
- Create several shallow parallel trenches across the bed.
The successful experiment did not install a compost-filled mushroom bed — it introduced colonized soil directly into existing orchard soil beneath established grass. A little mature, clean leaf mold can be mixed into extremely poor soil, but avoid creating a rich compost pile: Giant Puffball performed poorly in earlier nonsterile compost and waste experiments because it couldn't compete with other saprotrophic microorganisms.
11Transferring the Soil Spawn Outdoors
The published spawn was deposited ~7 cm (2¾ inches) below the surface and covered with soil.
- Water the bed lightly the day before if the soil is dry.
- Break the colonized soil spawn into small pieces inside its jar.
- Open the container outdoors.
- Create shallow trenches ~2½–3 inches deep, spaced ~12–16 inches apart.
- Distribute the spawn evenly through the trenches — avoid one concentrated hole.
- Cover it with the original soil and press the surface only lightly.
- Water gently to settle the soil, then mark the bed permanently.
- Record the date, amount of spawn, weather, and bed location.
Do not sterilize the outdoor soil. The goal is to introduce a vigorous culture into a stable natural soil environment.
A note on genetics — why one syringe is less certain
The published success used a mixture of two genetically different Giant Puffball isolates. DNA from the eventual mushrooms carried characteristics of both, suggesting that fusion or interaction between the isolates may have contributed to fruiting. A single cloned liquid culture may establish successfully, but its fruiting reliability is even less certain — worth knowing before you commit years to the bed.
12Maintaining the Bed
The bed should stay moderately moist, well aerated, protected from digging, covered by living grass or low vegetation, free from standing water, and exposed to normal seasonal cycles. Giant Puffball prefers moderate soil humidity, and natural fruiting is associated with substantial rainfall; the species isn't strongly heat-loving, and fruiting seems to depend more on soil moisture and season than on one narrow temperature.
Watering
In dry weather, check the soil beneath the surface and water when the upper two inches become dry. Apply slowly so it penetrates rather than running off, and let the surface begin drying between waterings. Don't keep the bed muddy or water heavily every day without checking. Natural rainfall is preferable to constant irrigation; during extended drought, provide just enough to prevent complete drying.
Mowing & fertilizer
Mow only when necessary, keeping blades several inches above the soil; don't drive heavy equipment over the bed, don't scalp the grass, remove heavy clipping piles, and stop mowing once a white fruiting structure appears. The research site was mowed only ~twice per year.
Do not routinely fertilize. Although Giant Puffball is associated with nutrient-rich soils, adding concentrated lawn fertilizer, manure, or compost doesn't guarantee fruiting and may encourage competing organisms.
13What Should Outdoor Colonization Look Like?
Visible mycelium may appear during the first several weeks and then apparently disappear. In the trial, small white underground structures appeared eight weeks after inoculation — then the visible mycelium disappeared and couldn't be found during inspections for the next three years, even though the fungus remained alive and eventually fruited.
Possible early signs
Fine white or ash-gray growth beneath the soil · small white compact knots · pale mycelial cords · colonized soil particles · dense gray-white growth several inches below the surface.
Do not repeatedly dig through the bed looking for mycelium. Disturbance can destroy the fine network you're trying to establish. The fungus may persist as visually undetectable hyphae for years.
14How Long Does It Take to Fruit?
This is a multi-year project.
The documented timeline
- March 2002: outdoor inoculation
- May 2002: small underground white structures observed
- 2002–2005: no visible mycelium or mushrooms
- September 2006: first aboveground puffball
- Aug & Sep 2007: two additional puffballs
The first mushroom appeared four years after inoculation, at the end of the fifth vegetation season.
So, realistically:
No mushroom during the first year is normal · no mushroom after two or three years does not prove failure · fruiting may require four or five growing seasons · a successfully colonized bed may never fruit · one bed may fruit while another nearby bed does not.
15The Fruiting Stage
Giant Puffballs develop directly from the soil as rounded white structures. Field observations found fruitbody enlargement continued ~5–11 days at a mean air temperature near 63.5°F, with high soil moisture appearing to be an important trigger. The first visible structure may look like a small white ball, a smooth white dome breaking through the grass, a rounded mass with no visible stem, or a firm white object partially buried in the soil.
Once a puffball appears
Stop mowing the area · keep pets and foot traffic away · don't cover it with mulch · don't spray it directly · maintain normal soil moisture · inspect it daily. Fruiting bodies can enlarge quickly, but size varies — the three cultivated mushrooms in the trial measured ~13, 14, and 16 cm in diameter.
16When Should I Harvest?
Harvest only while the entire fruiting body is young, firm, and white.
Identify Before You Eat — This Step Is Not Optional
Before considering any mushroom for culinary use: confirm it emerged from your established Giant Puffball bed, have it positively identified by an expert, cut it completely through the center, and examine the full interior. The interior must be solid, uniform, pure white, free from gills, free from any developing cap or stem, and free from yellow, olive, brown, purple, or black tissue.
Never consume a wild or outdoor-grown mushroom based on location or general appearance alone. Immature toxic mushrooms can resemble puffballs before their internal structures become visible. Poison Control advises against eating any wild mushroom unless it has been positively identified by an expert.
Harvest steps
Clean a sharp knife · cut through the base near the soil surface · avoid pulling up a large mass of soil · cut the mushroom completely in half · inspect the entire interior · discard it if any interior discoloration or mushroom structure is visible · refrigerate a correctly identified, completely white specimen promptly · cook thoroughly before eating.
Do not consume mature puffballs. Once the interior becomes yellow, olive, brown, or powdery, the tissue is converting into spores and is no longer in the proper culinary stage.
17Allowing a Puffball to Mature
A mature Giant Puffball can release an enormous amount of spores. Leaving one naturally developing fruiting body unharvested may help disperse spores around the established area, though it doesn't guarantee additional colonies.
When allowing one to mature
Keep it away from entrances and ventilation systems · don't kick, crush, or deliberately inhale the spore cloud · keep children and pets away · allow natural wind and weather to disperse the spores · avoid handling the dusty interior. The established underground mycelium is more important than the visible puffball for maintaining the bed.
18Additional Fruiting & Bed Longevity
Giant Puffball does not produce indoor-style flushes at predictable intervals. An established bed may produce one mushroom in a season, several in a season, skip one or more years, expand gradually into surrounding soil, or produce mushrooms several feet outside the original inoculation area. In the trial, later mushrooms appeared ~2½ and 5 meters beyond the original plot, and the authors estimated a minimum underground spread of ~one meter per year — so mark a larger area than the original bed and inspect nearby grass during late summer and early fall.
After harvesting
Replace disturbed soil gently · maintain ordinary moisture · avoid digging around the harvest point · don't soak the bed · continue normal low-disturbance maintenance.
Common Problems & What They Mean
Soil spawn colonizing very slowly. Slow growth is expected — the isolates grew ~4 mm/week on agar and needed ~6 weeks in sterilized soil at 77°F. Check incubation temperature, soil moisture, filter condition, culture distribution, and possible contamination. Don't push temperature substantially above 77°F to force growth.
Mycelium is gray instead of bright white. Light gray or ash-gray growth can be normal in soil spawn. Healthy growth is fine, organized, and associated with the soil. Green powder, black growth, pink colonies, slime, or foul odor is more concerning.
The jar is too wet. Wet soil restricts oxygen and encourages bacteria — signs include muddy texture, standing water, cloudy liquid, sour odor, and mycelium avoiding the saturated area. Don't open a sterile jar to add dry soil; prepare a replacement container with lower moisture.
The spawn colonized but disappeared after going outdoors. This doesn't necessarily mean the culture died — visible mycelium disappeared for ~three years in the successful trial before the first mushroom. Don't dig through the bed; keep protecting it and maintaining moderate moisture.
Small white knots appeared beneath the soil. Compact white structures may be encouraging — similar 8–12 mm structures appeared eight weeks after field inoculation in the plot that eventually fruited. They didn't immediately become aboveground mushrooms. Cover them gently and avoid further inspection.
No fruiting after one or two years. Expected. The only strongly documented artificial field fruiting took ~four years. Keep maintaining the site unless contamination, construction, flooding, or soil replacement has destroyed the bed.
One bed fruited but another didn't. The successful study had two nearby plots prepared the same way; only one fruited, with no simple mineral explanation. Local soil biology, moisture, competing organisms, and genetic compatibility may all matter.
The bed dries during summer. Water gradually over several sessions rather than flooding it. Maintain moisture beneath the surface while preserving air spaces in the soil.
The bed stays muddy. Reduce watering and improve surface drainage. Giant Puffball prefers moderate humidity but not standing water or compacted soil.
Other mushrooms appeared in the bed. An outdoor bed is a natural ecosystem — other fungi may appear even when Giant Puffball mycelium is present. Don't assume every mushroom is the intended species; identify each independently.
A white ball appeared but contains a mushroom shape inside. Do not eat it. A visible cap, stem, gills, internal outline, or gelatinous center means it is not a properly identified edible Giant Puffball.
The puffball turned yellow or olive inside. It has begun maturing into a spore mass and has passed the culinary stage. Leave it to mature naturally, or remove it carefully without inhaling the spores.
